تعیین مؤثرترین اسیدهای چرب مرتبط با چربی شیر گاوهای هلشتاین با استفاده از الگوریتم انتخاب ویژگی
آگاهی از تنظیم تولید چربی شیر جهت توسعه راهبردهای تغذیهای برای افزایش ارزش تغذیهای شیر، کاهش خروج انرژی از طریق آن و بهبود توازن انرژی گاوهای شیری حیاتی است. هدف از این پژوهش، تعیین مهمترین اسیدهای چرب مرتبط با چربی شیر گاوهای هلشتاین با استفاده از الگوریتم انتخاب ویژگی بود. الگوریتم انتخاب ویژگی یکی از روشهای دادهکاوی به منظور انتخاب بهترین و مؤثرترین فراسنجههای مرتبط برای پیشبینی هدف مورد نظر است. برای انجام این پژوهش از 3072 داده خام و سه روش مهم الگوریتم انتخاب ویژگی با شش مدل مختلف استفاده شد. داده با استفاده از نرمافزار WEKA واکاوی شد. نتایج نشان داد که روش رتبهبندی الگوریتم انتخاب ویژگی مناسبترین روش برای انتخاب مهمترین اسیدهای چرب مرتبط با چربی شیر با استفاده از رگرسیون لجستیک و طبقهبندی درختی با کمترین میزان خطا است. بر این اساس، مهمترین اسیدهای چرب مرتبط با ساخت چربی شیر، ایکوزا تری انوئیک اسید (20:3C 3 -n)، ایکوزا دی انوئیک اسید (20:2C 6 -n)، مجموع کل اسیدهای چرب ترانس 16:1C، ترانس-10 16:1C، ترانس-6، 7، 8 16:1C، مجموع کل اسیدهای چرب غیراشباع با یک پیوند دوگانه ترانس، ترانس-11، سیس-15 18:2C، مجموع کل اسیدهای چرب ترانس 18:1C، آدرنیک اسید (20:4C 6 -n) و سیس-12 18:1C تعیین شدند. الگوریتم انتخاب ویژگی اثر اسیدهای چربی به غیر از ایزومر ترانس-10، سیس-12 اسید لینولئیک مزدوج و ایزومر ترانس-10 18:1C را بر ساخت چربی شیر مؤثر نشان داد که ضروری است اهمیت زیستشناختی آنها در پژوهشهای آینده مورد سنجش قرار بگیرد.

ایجاد درخت ژنی با استفاده از واگرایی کولبک-لیبلر روی ژنهای موثر بر تولید شیر در گاو شیری
نظریه اطلاعات، شاخهای از ریاضیات است که با مهندسی ارتباطات، زیستشناسی و پزشکی همپوشانی دارد. هدف از بررسی حاضر ارائه روشی جهت خوشهبندی تعدادی از ژنهای موثر روی تولید شیر در گاو شیری با استفاده از الگوریتمی متکی بر واگرایی کولبک - لیبلر بود. در این پژوهش بعد از استخراج توالی DNA ژن و اگزونهای موثر بر تولید شیر در گاو شیری، فراسنجه آنتروپی در مراتب یک تا چهار برای هر ژن و اگزونهای هر ژن محاسبه شد. جهت استخراج فاصله میان ژنها از یکدیگر، از واگرایی کولبک - لیبلر در سه روش مختلف استفاده شد. روشهای اول و دوم مبتنی بر همترازی ولی روش سوم غیر مبتنی بر همترازی و بر پایه آنتروپی نسبی ژنها بود. نتایج هر سه روش واگرایی کولبک - لیبلر روی توالی DNA ژنها و اگزونها با استفاده از هفت روش معمولSingle ،Complete ،Average ،Weighted ، Centroid، Medianو KMeansخوشهبندی شدند. تجمیع نتایج هر خوشهبندی که با الگوریتم AdaBoost انجام شد، و خود نوعی درخت ژنی را تداعی کرد، نشان داد که روش سوم، خوشهبندی معقولی را از نظر زیستی برای مجموعهای از ژنها حاصل نمود چرا که با نتایج حاشیهنویسی ژنومی ژنهای حاصل ازGeneMANIA مطابقت داشت. این اعتقاد وجود دارد که روش ارائه شده برای ایجاد درخت ژنی میتواند با سایر روشهای متکی بر توالی DNA ژنها جهت خوشهبندی مجموعهای از ژنها، رقابت نماید و لذا میتواند در گروهبندی ژنهای سایر گونهها نیز بکار رود.

شناسایی مناطق ژنومی مرتبط با صفات تولیدمثلی در گوسفند بلوچی با استفاده از نشانگرهای با تراکم بالا
در این تحقیق، یک پویش ژنومی برای شناسایی جایگاههای مؤثر بر صفات تولیدمثلی در گوسفند بلوچی انجام شد. به این منظور، نمونهبرداری خون از 96 رأس میش همراه با دادههای فنوتیپی مربوط به صفات تولیدمثلی شامل مجموع وزن همزادان متولد شده، میانگین وزن همزادان در تولد، مجموع وزن همزادان شیرگیری شده، میانگین وزن همزادان شیرگیری شده و تعداد برههای زنده در شش ماهگی در چهار نوبت زایش بدست آورده شد. پس از استخراج DNA، نمونهها با استفاده از تراشههایSNP گوسفندی (50K) تعیین ژنوتیپ شدند. تجزیه مطالعه ارتباط ژنومی با استفاده از نرمافزار PLINK در مدلهای رگرسیون خطی و لجستیک شامل اثرات SNPها و عوامل ثابت جنس و نوبت زایش انجام شد. در مجموع، هشت نشانگر معنیدار در سطح کروموزومی روی کروموزومهای 1، 4، 10، 15 و 17 مرتبط با صفات تولیدمثلی مورد مطالعه شناسایی شد (05/0>P). بررسی ژنها و QTLها در این مناطق نیز نشاندهنده وجود ژنها و QTLهای مؤثر بر صفات رشد و تولیدمثلی بود. از نتایج این تحقیق میتوان برای کشف واریانتهای مسبب صفات تولیدمثلی در برنامههای اصلاح نژادی گوسفند استفاده نمود.

اثر همخونی بر میانگین افزایش وزن روزانه و نسبت کلیبر در گوسفندان استان گیلان
هدف از این پژوهش بررسی اثر همخونی روی صفات رشد گوسفندان استان گیلان بود. صفات مورد مطالعه شامل میانگین افزایش وزن روزانه از تولد تا سهماهگی (ADG1)، از تولد تا ششماهگی (ADG2) و از سهماهگی تا ششماهگی (ADG3) و همچنین نسبتهای کلیبر منطبق با افزایش وزنهای روزانه فوقالذکر (بهترتیب KR1، KR2 و KR3) بود. در این پژوهش، از اطلاعات شجره و دادههای جمعآوری شده به وسیله سازمان جهاد کشاورزی استان گیلان در طی سالهای 1373 تا 1390 استفاده شد. برای بررسی اثر همخونی بر صفات مورد مطالعه (میزان تغییر صفات بهازای یک درصد افزایش همخونی) از رویهReg نرم افزارSAS به تفکیک برای تیپ تولد، جنس و کل جمعیت استفاده شد. اثر همخونی بر تمامی صفات به جز ADG1 مثبت بود. ضریب رگرسیون ADG1، ADG2 و KR2 به ترتیب 138/0- گرم، 101/0 گرم و 0053/0 بودند (01/0P<). ضریب رگرسیون ADG3 و KR3 به ترتیب برابر 071/0 و 0096/0 گرم بودند (05/0P<) و همچنین ضریب رگرسیون KR1 برابر با 0023/0 بود و روند معنیداری نداشت. اگر چه تاکنون افت همخونی در عملکرد صفات رشد گوسفندان استان گیلان مشاهده نشده است اما استفاده از سیستم جفتگیری طراحی شده در گله میتواند یک روش مناسب برای حفظ سطح همخونی تحت کنترل باشد.

اثر افزودن اسانس نعناع فلفلی و آویشن شیرازی به جیره آغازین بر عملکرد گوساله های هلشتاین
هدف از انجام این تحقیق، بررسی اثر افزودن اسانسهای نعناع فلفلی و آویشن شیرازی به جیره آغازین گوسالههای شیرخوار هلشتاین بر میزان مصرف خوراک، عملکرد رشد، سن از شیرگیری، قابلیت هضم مواد مغذی و قوام مدفوع بود. در این آزمایش 36 راس گوساله هلشتاین بهطور تصادفی به سه جیره غذایی با 12 رأس گوساله (6 راس نر و 6 راس ماده) اختصاص داده شدند. جیرههای غذایی شامل 1) خوراک آغازین بدون افزودنی (شاهد) 2) خوراک آغازین حاوی 2/0 درصد اسانس نعناع فلفلی و 3) خوراک آغازین حاوی 2/0 درصد اسانس آویشن شیرازی بود. گوسالهها در جایگاههای انفرادی نگهداری شده و به آب و خوراک دسترسی آزاد داشتند. از شیرگیری در زمان مصرف روزانه یک کیلوگرم ماده خشک در دو روز متوالی انجام شده و آزمایش تا دو هفته بعد از شیرگیری ادامه یافت. میزان مصرف ماده خشک به صورت روزانه اندازهگیری شده و وزنکشی در سنین یکماهگی، 2 ماهگی، زمان از شیرگیری و پایان آزمایش انجام شد. قوام ظاهری مدفوع به صورت یک روز در میان اندازهگیری شد. تفاوت معنیداری بین تیمارها از نظر سن از شیرگیری و میزان خوراک مصرفی وجود نداشت. گوسالههای مصرفکننده جیره حاوی اسانس نعناع فلفلی، افزایش وزن روزانه و بازده مصرف خوراک بیشتری نسبت به تیمار شاهد داشتند (05/0P<). تیمارهای آویشن و نعناع باعث سوددهی بیشتر در زمان از شیرگیری نسبت به گروه شاهد شدند. نتایج این آزمایش نشان داد افزودن اسانس نعناع به جیرههای آغازین باعث بهبود بازده مصرف خوراک و در نهایت افزایش سود ناخالص پرورش گوساله شد.

اثر جایگزینی یونجه با غلاف سوبابل (Leucaena leucocephala ) بر قابلیت هضم، تخمیر آزمایشگاهی و تجزیهپذیری (in situ) در گاو و گاومیش
این آزمایش به منظور بررسی تاثیر جایگزینی یونجه با غلاف سوبابل (0، 9 و 18 درصد ماده خشک) بر قابلیت هضم، تخمیر آزمایشگاهی و تجزیهپذیری شکمبهای در گاو و گاومیشفیستوله شده انجام شد. تخمیر جیرههای آزمایشی با تولید گاز، قابلیت هضم با تلی و تری و تجزیهپذیری با روش کیسههای نایلونی تعیین شد (چهار تکرار برای هر جیره) و دادهها در قالب طرح کرتهای خرد شده تجزیه شدند. قابلیت هضم ماده خشک و الیاف نامحلول در شوینده خنثی و اسیدی در جیرههای حاوی سطوح مختلف غلاف سوبابل بین گاو و گاومیش تفاوتی نداشت (05/0<P)، اما پتانسیل تولید گاز در جیرههای حاوی غلاف سوبابل کمتر از جیره شاهد بود (05/0>P). جایگزینی یونجه با غلاف سوبابل اثری بر نرخ تولید گاز، تولید توده میکروبی و دیگر فراسنجههای تخمیری در گاو و گاومیش نداشت (05/0P>). بالاترین مقدار تجزیه دیواره سلولی مربوط به تیمار شاهد (در گاومیش، 9/23 درصد و در گاو، 9/35 درصد) و کمترین مقدار مربوط به تیمار حاوی 18 درصد غلاف سوبابل (در گاومیش، 1/64 درصد و در گاو، 3/64 درصد) بود (05/0>P). جایگزینی یونجه با غلاف سوبابل، فراسنجههای تجزیهپذیری شکمبهای در گاو و گاومیش را تحت تاثیر قرار نداد (05/0P>). با توجه به نتایج، جایگزینی علوفه یونجه با غلاف سوبابل در جیره گاو و گاومیش، بر قابلیت هضم و تولید گاز تاثیر نداشت، اما تجزیهپذیری شکمبهای را بهبود داد. بنابراین شاید بتوان 50 درصد غلاف سوبابل را به جای یونجه در تغذیه گاو و گاومیش استفاده کرد.

اثر سطوح مختلف ملاس و ویناس نیشکر در جیره غذایی بر فراسنجههای تولید گاز در شرایط آزمایشگاهی
این مطالعه جهت تعیین ارزش و مقایسه فراسنجههای تولید گاز ملاس و ویناس در قالب طرح کاملاً تصادفی با پنج تیمار و شش تکرار انجام شد. جهت تهیه مایع شکمبه از سه رأس گوسفند نر بالغ فیستولدار استفاده شد. جیرههای آزمایشی شامل: جیره 1) جیره پایه +20 درصد ملاس + صفر درصد ویناس (شاهد)، جیره 2) جیره پایه + 15 درصد ملاس + 5 درصد ویناس، جیره 3) جیره پایه + 10 درصد ملاس + 10 درصد ویناس، جیره 4) جیره پایه + 5 درصد ملاس + 15 درصد ویناس، و جیره 5) جیره پایه + صفر درصد ملاس + 20 درصد ویناس بودند. بر اساس نتایج به دست آمده پتانسیل تولید گاز، نرخ گاز تولید شده، انرژی قابل متابولیسم، قابلیت هضم ماده آلی و مقدار اسیدهای چرب کوتاه زنجیر در ملاس بطور معنیداری بیشتر از ویناس بود (05/0P<). عامل تفکیک، تولید توده میکروبی و راندمان تولید توده میکروبی در سطح 10 درصد ویناس کاهش معنیداری نسبت به شاهد نشان داد (05/0P<). حجم تولید گاز در سطح 15 درصد ویناس در تمامی ساعات انکوباسیون نسبت به گروه شاهد به طور معنیداری بیشتر بود (05/0P<). مقدار انرژی قابل متابولیسم، قابلیت هضم ماده آلی و مقدار اسیدهای چرب کوتاه زنجیر در در سطح 15 درصد ویناس افزایش معنیداری نسبت به شاهد نشان داد(05/0P<). به طور کلی جایگزینی ویناس با ملاس به نسبت 15 درصد در جیرههای غذایی، باعث افزایش ارزش غذایی جیرهها میشود.

بررسی روند تجزیه بقایای سموم فوزالون و دیازینون در علف یونجه تازه، سیلو شده و انبارشده به صورت خشک
آزمایشی با هدف مقایسه روند تجزیه بقایای سموم فوزالون و دیازینون در علف یونجه تازه، سیلو شده و انبار شده بهصورت خشک در شرایط مزرعه انجام شد. بخشی از مزرعه یونجه جدا و به کرتهای مساوی تقسیم و با سموم فوزالون و دیازینون سمپاشی شد. نمونههایی از علف یونجه تازه، سیلو شده و خشک شده با فواصل زمانی مشخص به ترتیب تا زمانهای 15 روز، 14 و 24 هفته بعد از سمپاشی، گرفته شد. بقایای سموم با روش گازگروماتوگرافی جرمی (GC-Mass) تعیین شدند. مقدار فوزالون و دیازینون در روز پانزدهم پس از سمپاشی در علف یونجه تازه به ترتیب برابر با 13/2±33/57 و 51/1±75/24 میلیگرم در هر کیلوگرم ماده خشک بود. زمان تجزیه شدن50 درصد فوزالون و دیازینون در علف یونجه تازه به ترتیب برابر با 34/4 و 62/3 روز بود. هیچگونه بقایایی از دیازینون در 14 هفته بعد از انبار کردن علف یونجه به صورت خشک مشاهده نشد، ولی مقدار فوزالون در علف یونجه خشک تا 24 هفتگی برابر 32/3 میلیگرم بود. همچنین در اثر سیلو کردن، هیچگونه بقایایی از دیازینون و فوزالون بهترتیب در هفتههای 12 و 14 نمونهگیری مشاهده نشد. نتایج کلی آزمایش نشان داد فوزالون نسبت به دیازینون از قابلیت تجزیه کمتری در هر سه علف یونجه تازه، سیلوشده و انبارشده بهصورت خشک برخوردار بوده و این دو سم میتوانند بهطور نسبی تا مدتها در علف یونجه باقی بمانند. همچنین سیلوکردن نسبت به انبارکردن علف یونجه به صورت خشک، تأثیر بیشتری بر کاهش باقیمانده سموم با گذشت زمان داشت.
