بررسی چند شکلی ژن های گیرنده لپتین و گیرنده ملانوکورتین4 با استفاده از نشانگر PCR-SSCP در گاوهای بومی استان گیلان

چکیده

تحقیق حاضر جهت بررسی چندشکلی ژن­های گیرنده لپتین (LEPR) و گیرنده ملانوکورتین 4 (MC4R) در گاوهای بومی استان گیلان انجام شد. بدین منظور از 96 رأس گاو بومی استان گیلان به طور تصادفی خونگیری به عمل آمد. پس از استخراج DNA با استفاده از روش نمکی بهینه یافته، واکنش زنجیره ­ای پلیمراز (PCR) برای تکثیر قطعه 269 جفت بازی اگزون 2 ژن LEPR و قطعه 213 جفت بازی از اگزون 1 ژن MC4R انجام شد. چندشکلی فضایی تک­رشته­­ای (SCCP) محصولات RCR با استفاده از ژل اکریل آمید و رنگ­آمیزی نیترات نقره به دست آمد. برای ژن LEPR در نمونه مورد مطالعه 3 الگوی باندی AA، AB وAC  و برای ژن MC4R، 6 الگوی باندی AA، AB، AC، AD، AE، AF و AG مشاهده شد. فراوانی­های ژنوتیپی برای ژن LEPR به ترتیب 062/0، 615/0 و 323/0 و برای ژنوتیپ ­های ژن MC4R، 074/0، 21/0، 253/0، 158/0، 105/0، 105/0 و 095/0 بود. تجزیه و تحلیل ژنتیک جمعیت نشان داد که ژن­­های LEPR و MC4R در تعادل هاردی-وینبرگ نبودند، اما هر دو ژن از تنوع ژنتیکی و چندشکلی بالایی برخوردار بودند.

چکیده [English]

The experiment was concluded to study the genetic polymorphisms for Exon 2 leptinreceptor (LEPR) and Exon 1Melanocortin4 receptor (MC4R) genes in native cattle in Guilan province. In order to blood samples were collected randomly from 96 native cattle in Guilan province. After extracting DNA by salting out method, PCR-SSCP was used to amplify a fragment of 261 bp from exon 2 LEPR gene and a fragment of 213 bp from exon 1 MC4R gene. In the sample of study, three patterns AA, AB and AC for LEPR gene and six patterns AA, AC, AD, AE, AF and AG for MC4R gene were observed. Frequencies of genotype were 0.062, 0.615 and 0.323 for LEPR gene and 0.074, 0.21, 0.253, 0.158, 0.105, 0.105 and 0.095 for MC4R gene respectively. Population genetic analysis showed that LEPR and MC4R genes were out of Hardy-Weinberg equilibrium but both genes were highly polymorphic and had great genetic diversity.
