بررسی اسیدیته دستگاه گوارش و خصوصیات استخوان درشتنی مرغان تخمگذار تغذیه شده با مکملهای آنتیبیوتیک، اسید آلی، پروبیوتیک و پریبیوتیک
این مطالعه به منظور بررسی اثر استفاده از مکملهای آنتیبیوتیک، اسید آلی، پروبیوتیک و پریبیوتیک بر اسیدیته بخشهای مختلف دستگاه گوارش، مواد معدنی پلاسمای خون و خصوصیات استخوان درشتنی مرغان تخمگذار انجام شد. آزمایش در قالب طرح کاملاً تصادفی با 160 قطعه مرغ تخمگذار سویه های-لاین (W-36) از سن 32 تا 42 هفتگی با 5 تیمار، 4 تکرار و 8 قطعه مرغ تخمگذار در هر تکرار انجام گرفت. تیمارهای آزمایشی شامل: 1- تیمار شاهد (جیره پایه، بدون افزودنی)، 2- جیره پایه + 150 گرم در تن آنتیبیوتیک اکسی تتراسایکلین، 3- جیره پایه + 3 کیلوگرم در تن مکمل اسید آلی اُرگاسید، 4- جیره پایه + 50 گرم در تن پروبیوتیک پروتوکسین و 5- جیره پایه + 2 کیلوگرم در تن پریبیوتیک مانان الیگوساکارید بودند. نتایج نشان داد اسیدیته چینهدان، دئودنوم و ژژنوم در اثر استفاده از مکمل اسید آلی بطور معنیداری کاهش یافت (05/0P<). غلظت مواد معدنی پلاسمای خون (کلسیم، فسفر، منیزیم و آهن) و همچنین کلسیم و فسفر استخوان درشتنی تحت تأثیر تیمارهای آزمایشی قرار نگرفت (05/0<P). خاکستر استخوان درشتنی و قطر خارجی آن به وسیله مکمل اسید آلی افزایش معنیداری پیدا کرد (05/0P<). همچنین استفاده از مکملهای اسید آلی و پروبیوتیک باعث بهبود استحکام و مقاومت استخوان درشتنی در برابر ضربه شد. نتایج بدست آمده از این مطالعه نشان میدهد استفاده از مکمل اسید آلی میتواند اثرات مفیدی بر خصوصیات استخوان درشتنی مرغان تخمگذار داشته باشد.

بررسی اثر متقابل بین تعادل کاتیون-آنیون جیره و فیتاز میکروبی بر عملکرد، خصوصیات لاشه و پاسخ ایمنی جوجه های گوشتی
در این تحقیق به منظور بررسی آثار اصلی و متقابل سطوح مختلف تعادل کاتیون-آنیون جیره و آنزیم فیتاز میکروبی بر عملکرد جوجه، خصوصیات لاشه و پاسخ ایمنی، از 1200 قطعه جوجه گوشتی نر سویه تجارتی راس استفاده شد. آزمایش فاکتوریل 3×4 با چهار سطح تعادل کاتیون- آنیون جیره (200، 225، 250 و 275 میلیاکیوالان در کیلوگرم جیره) و سه سطح آنزیم فیتاز میکروبی در جیره (صفر، 500 و 750 واحد فیتازی در کیلوگرم جیره) استفاده شد. نتایج نشان داد آثار متقابل بین تعادل کاتیون- آنیون جیره و آنزیم فیتاز باعث تفاوت آماری معنیدار (05/0> P) بر صفات عملکردی (خوراک مصرفی، ضریب تبدیل غذایی، افزایش وزن، شاخص تولید و وزن بدن جوجههای گوشتی در سن 42 روزگی) شد. آثار متقابل بین تعادل کاتیون-آنیون جیره و فیتاز میکروبی موجب عدم تفاوت معنیدار (05/0 < P) بین تیمارها از نظر خصوصیات لاشه (وزن لاشه، رانها، سینه، پشت + گردن، کبد، قلب، طحال، سنگدان، بورس فابرسیوس و چربی حفره شکمی و همچنین وزن و طول قسمتهای مختلف روده باریک جوجههای گوشتی) شد. آثار متقابل بین تعادل کاتیون- آنیون جیره و فیتاز میکروبی باعث تفاوت آماری معنیدار (05/0 > P) از نظر سیستم ایمنی جوجههای گوشتی در دو سن 24 و 34 روزگی شد. به طور کلی، نتایج حاصل از پژوهش حاضر نشان داد که تنوع پاسخ در عملکرد و سیستم ایمنی جوجههای گوشتی به مکمل فیتاز در جیره تا حدی میتواند تحت تأثیر تفاوت در سطوح تعادل کاتیون-آنیون جیره باشد.

اثر مکمل جیره ای پری بیوتیک و اسیدهای آلی بر عملکرد، خصوصیات لاشه و ریختشناسی روده جوجه های گوشتی
تحقیق حاضر به منظور بررسی اثر پریبیوتیک سافمانان و اسید آلی لیپتول HPA بر عملکرد، خصوصیات لاشه و مورفولوژی روده کوچک جوجه گوشتی انجام شد. در این آزمایش تعداد 160 قطعه جوجه گوشتی یکروزه سویه راس 308 در قالب طرح کاملاً تصادفی با 4 تیمار و 4 تکرار و تعداد 10 قطعه جوجه نر برای هر تکرار در یک دوره 42 روزه استفاده شدند. تیمارهای آزمایشی شامل جیره شاهد و جیرههای حاوی 1/0 درصد پریبیوتیک، 1/0 درصد اسید آلی و 1/0 درصد مخلوطی از اسید آلی و پریبیوتیک بودند. مصرف خوراک، افزایش وزن بدن و ضریب تبدیل غذا بهصورت دورهای و شاخص تولید، ویژگیهای لاشه و صفات مورفولوژیکی روده در 42 روزگی اندازهگیری شدند. در کل دوره در بین تیمارهای آزمایشی تفاوت معنیداری از نظر مصرف خوراک مشاهده نشد. بیشترین افزایش وزن روزانه جوجهها در جیره حاوی اسید آلی مشاهده شد (05/0>P). همچنین بهترین ضریب تبدیل غذایی و شاخص تولید مربوط به جیره حاوی اسیدهای آلی و بدترین آنها مربوط به جیره شاهد بود (05/0>P). بیشترین درصد لاشه در جیره حاوی اسید آلی و بیشترین درصد ران و سینه در جیره حاوی پریبیوتیک مشاهده شد (05/0>P). همچنین بیشترین ارتفاع ویلی مربوط به جیره حاوی اسید آلی و کمترین آن مربوط به تیمار شاهد بود (05/0P<). عمق کریپت در بین جیرههای آزمایشی معنیدار نشد و کمترین ضخامت لایه ماهیچههای روده مربوط به تیمار حاوی اسید آلی بود. نتایج بهدستآمده از این تحقیق نشان داد جیره دارای 1/0 درصد اسید آلی نسبت به سایر جیرهها موجب بهبود عملکرد، خصوصیات لاشه و فراسنجههای روده جوجههای گوشتی شد.

مقایسه پیشرفت ژنتیکی در تولید شیر گاو هلشتاین در ایران ناشی از آزمون نتاج و انتخاب ژنومی و اثر تعداد گاو نر جوان بر آن
در این تحقیق رشد ژنتیکی حاصل از یک برنامه آزمون نتاج مطابق با مشخصات جمعیتی گاو هلشتاین در ایران و یک برنامه انتخاب ژنومی معادل از نظر تعداد والدین نر و ماده مورد نیاز مقایسه و اثر تعداد گاوهای نر جوان بر رشد ژنتیکی در این دو برنامه بررسی شد. هدف انتخاب صفت تولید شیر بود. رشد ژنتیکی حاصل از انتخاب چهار مسیری با استفاده از روش جریان ژن و در یک افق 150 ساله برای صفت تولید شیر برآورد شد. نتایج نشان داد که آزمون نتاج و انتخاب ژنومی علاوه بر صحت انتخاب از نظر شدت انتخاب و نیز فاصله نسل متفاوتند. رشد ژنتیکی سالانه در آزمون نتاج برابر 7/114 و در انتخاب ژنومی برابر 7/173 کیلوگرم بود. نتایج حاصل از تغییر تعداد گاو نر جوان تحت آزمون از 50 تا 300 رأس نشان داد، رشد ژنتیکی در آزمون نتاج بین 11/106 تا 53/117 کیلوگرم و در انتخاب ژنومی بین 95/160 تا 38/185 کیلوگرم متغیر است. در هر دو برنامه رشد ژنتیکی نسبت به افزایش تعداد گاو نر جوان هنگامی که تعداد آن کمتر از 100 رأس بود، حساسیت بیشتری داشت. بطور کلی این تحقیق نشان داد که رشد ژنتیکی حاصل از انتخاب ژنومی در مقایسه با آزمون نتاج به دلیل فاصله نسل کوتاهتر، میتواند بیش از 50 درصد برتری داشته باشد. اما این برتری تحت تاثیر تعداد گاو نر جوان قرار داشته و در ازای 75 رأس به کمترین مقدار خود می رسد.

بررسی چند شکلی ژن های گیرنده لپتین و گیرنده ملانوکورتین4 با استفاده از نشانگر PCR-SSCP در گاوهای بومی استان گیلان
تحقیق حاضر جهت بررسی چندشکلی ژنهای گیرنده لپتین (LEPR) و گیرنده ملانوکورتین 4 (MC4R) در گاوهای بومی استان گیلان انجام شد. بدین منظور از 96 رأس گاو بومی استان گیلان به طور تصادفی خونگیری به عمل آمد. پس از استخراج DNA با استفاده از روش نمکی بهینه یافته، واکنش زنجیره ای پلیمراز (PCR) برای تکثیر قطعه 269 جفت بازی اگزون 2 ژن LEPR و قطعه 213 جفت بازی از اگزون 1 ژن MC4R انجام شد. چندشکلی فضایی تکرشتهای (SCCP) محصولات RCR با استفاده از ژل اکریل آمید و رنگآمیزی نیترات نقره به دست آمد. برای ژن LEPR در نمونه مورد مطالعه 3 الگوی باندی AA، AB وAC و برای ژن MC4R، 6 الگوی باندی AA، AB، AC، AD، AE، AF و AG مشاهده شد. فراوانیهای ژنوتیپی برای ژن LEPR به ترتیب 062/0، 615/0 و 323/0 و برای ژنوتیپ های ژن MC4R، 074/0، 21/0، 253/0، 158/0، 105/0، 105/0 و 095/0 بود. تجزیه و تحلیل ژنتیک جمعیت نشان داد که ژنهای LEPR و MC4R در تعادل هاردی-وینبرگ نبودند، اما هر دو ژن از تنوع ژنتیکی و چندشکلی بالایی برخوردار بودند.

مقایسه بیان ژن های لاکتوفرین و TLR4 در سلول های سوماتیک شیر گاو هلشتاین، بومی گیلان و آمیختههای حاصل از آنها
هدف تحقیق حاضر مقایسه بیان ژنهای لاکتوفرین و TLR4 در سلولهای سوماتیک شیر گاو و بررسی تفاوتهای ژنتیکی و آمیختهگری بر بیان ژنهای سیستم ایمنی و همچنین اثرات ورم پستان بر بیان ژنهای مذکور است. RNA کل از سلولهای سوماتیک شیر تازه 3 گاو سالم هلشتاین، 3 گاو بومی گیلان و 3 گاو از آمیختههای نسلF1 حاصل از آنها در مرکز اصلاح نژاد گاو بومی گیلان و 3 گاو هلشتاین دارای علائم خفیف ورم پستان بالینی استخراج شد. پس از ساخت cDNA، بیان ژنها با استفاده از Real-time PCR نسبت به ژن مرجع GAPDH اندازهگیری شد. نتایج نشان داد درحالیکه متوسط بیان هر دو ژن در آمیختهها بهطور معنیداری بالاتر از والدین بود (05/0P>)، انحراف استاندارد بیان ژن لاکتوفرین در جمعیت آمیختهها نیز بالا بود. بیان ژن TLR4 در گاوهای هلشتاین با علائم ورم پستان نسبت به هلشتاین بدون ورم پستان بهطور معنیداری بیشتر بود (05/0P>)، اما تفاوت معنیداری در بیان ژن لاکتوفرین بین گاوهای هلشتاین سالم و هلشتاین با ورم پستان مشاهده نشد (05/0P>). این مطالعه ضمن نشان دادن عدم تفاوت معنیدار بیان هر دو ژن مورد بررسی در گاوهای هلشتاین و بومی، اثر آمیختهگری را بر بیان ژنهای مرتبط با مقاومت ژنتیکی به خوبی نشان داد.

مدلسازی راندمان انرژی در تولید مرغ گوشتی به کمک رویکرد شبکههای عصبی مصنوعی پرسپترون (مطالعه موردی: استان اردبیل)
سامانههای تولیدی در صنایع زیستی بر مدیریت منابع و تبدیل صور مختلف انرژی استوار است. این پژوهش به بررسی و مدلسازی غیرپارامتریک انرژی تولید مرغ گوشتی پرداخته است. نمونههای مورد بررسی، مشتمل بر 70 تولیدکننده مرغ گوشتی استان اردبیل بود که بهطور تصادفی از جامعه آماری منطقه انتخاب شدند. در این مطالعه، معادل انرژی مصرفی در صنعت مذکور برآورد شد و سپس شاخصهای انرژی محاسبه شد. سپس با استفاده از مدلهای شبکه عصبی مصنوعی میزان معادل انرژی ستانده و عملکرد سیستم، مدلسازی و برآورد شد. بر اساس نتایج حاصل از پژوهش، میزان کل انرژی معادل نهاده و ستانده در تولید مرغ گوشتی به ترتیب در حدود 79/153 و 45/27 گیگاژول بر 1000 قطعه مرغ محاسبه شد. بیشترین انرژی نهاده مصرفی در تولید مرغ گوشتی منطقه متعلق به سوخت فسیلی با 48/61 درصد از کل معادل انرژی بود. بر اساس نتایج شبکه عصبی مصنوعی بهترین ساختار برای مدلسازی انرژی مصرفی تولید مرغ گوشتی ساختار 2-14-5 با 5 ورودی، یک لایه مخفی با 14 نرون و یک لایه خروجی با دو پارامتر خروجی تخمین زده شد. ضرایب تبیین مناسبترین ترکیب وزن برای تخمین معادل انرژی تولید گوشت مرغ و کود بستر به ترتیب 93 و 91 درصد برای دادههای آزمون و 98 و 95 درصد برای دادههای اعتبارسنجی بهدست آمد. در ارزیابی تاثیرپذیری خروجی از نهادههای ورودی، سوخت فسیلی بالاترین حساسیت را در بین نهادههای تولیدی از خود نشان داد که لزوم بازنگری در منابع انرژی را بیش از پیش آشکار میسازد.
