استخراج شبکه بین زیست نشانگرهای ترانسکریپتومی ورم پستان گاو شیری ناشی از باکتری استافیلوکوکوس اورئوس با استفاده از ژنوم انسان
ورم پستان یکی از بیماریهای عفونی غدد پستانی در گاوهای شیری است که در چند دهه اخیر، به عنوان یکی از مهمترین بیماریهایی که منجر به ضرر اقتصادی در صنعت گاو شیری میشود، مورد توجه زیادی قرار گرفته است. وراثتپذیری ورم پستان پایین است و لذا مدیریت اصلاح نژادی آن کار سختی به شمار میرود. با توجه به اهمیت بیماری ورم پستان در مدیریت واحدهای پرورش گاو شیری و ساز و کار مولکولی پیچیده درگیر در این بیماری، استخراج شبکه بین زیستنشانگرهای ترانسکریپتومی ورم پستان با استفاده از دادههای امیکس موجود در گونههایی مثل انسان، که حاشیهنویسی ژنومی قابل اعتمادی دارند، معقول به نظر میرسد. در این پژوهش با استفاده از برازش یک مدل خطی مشابه روی هر ژن یک آزمایش ریزآرایه DNA انجام شده ناشی از استافیلوکوکوس اورئوس بر پستان گاو شیری، فهرستی از ژنهای متفاوت بیان شده یا زیستنشانگرهای ترانسکریپتومی گاوی بهدست آمدند. نتایج نشان داد ژنهای رمز کننده سایتوکینها و کایموکینهای CXCL5، CCL2،CXCL2 ، CXCL3، CXCL8 و SAA3 در اثر ورم پستان ناشی از استافیلوکوکوس اورئوس، بیان متفاوتی داشتند. در این پژوهش با نگاشت مجموعه ژنهای متفاوت بیان شده روی ژنوم انسان، ژنهای ارتولوگ انسان و گاو بهدست آمدند. شبکه زیستنشانگری بهدست آمده از ژنهای ارتولوگ انسان نشان داد که نشانگر APP (Amyloid beta precursor protein) بیشترین درجه ارتباط را با سایر نشانگرهای ترانسکریپتومی دارد. به نظر میرسد از نتایج این پژوهش بتوان در برنامههای اصلاح نژادی و تهیه دارو برای درمان ورم پستان استفاده نمود.

اثر افزودن سطوح مختلف ساپونین، اسید گالیک و اسید تانیک بر کینتیک تخمیر شکمبه ای در شرایط برون تنی
این مطالعه به منظور بررسی اثر سطوح مختلف (5/2، 5 و 10 درصد ماده خشک) ساپونین، اسید گالیک و اسید تانیک بر فراسنجههای تولید گاز و قابلیت هضم برونتنی ماده خشک و ماده آلی با استفاده از روش تولید گاز و کشت ثابت انجام گرفت. نتایج نشان داد که بین تیمارهای آزمایشی از نظر پتانسیل و نرخ تولید گاز اختلاف معنیداری وجود داشت (05/0>P). تیمار شاهد و تیمارهای دارای ساپونین به ترتیب دارای بالاترین و پایینترین پتانسیل تولید گاز بودند. به هر حال، افزودن سطوح مختلف ساپونین، اسید گالیک و اسید تانیک به جیره پایه سبب کاهش قابلیت هضم ماده آلی، انرژی قابل متابولیسم و غلظت اسیدهای چرب کوتاه زنجیر شد. از این نظر، تیمار دارای 10 درصد اسید گالیک پایینترین مقدار قابلیت هضم ماده آلی، انرژی قابل متابولیسم و غلظت اسیدهای چرب کوتاه زنجیر را داشت. بالاترین مقدار غلظت نیتروژن آمونیاکی مربوط به تیمار شاهد (76/10 میلیگرم در دسیلیتر) و پایینترین مقدار مربوط به تیمار دارای 10 درصد اسید تانیک (79/5 میلیگرم در دسیلیتر) بود. بین تیمارها از نظر بازده تولید گاز، عامل تفکیک و تولید توده میکروبی تفاوت معنیداری وجود داشت (05/0P<). به طور کلی، نتایج این مطالعه نشان داد که استفاده از سطوح مختلف ساپونین، اسید گالیک و اسید تانیک مورد استفاده اثر قابل ملاحظهای بر فرآیندهای تخمیر شکمبهای در شرایط آزمایشگاهی نداشت.

اثر استفاده از منابع مختلف نیتروژن بر گوارشپذیری و تعادل نیتروژن در برههای نر نژاد مهربان
این پژوهش به منظور مقایسه اثر منابع مختلف نیتروژن بر روند آزادسازی، گوارشپذیری و متابولیسم نیتروژن در برههای نر نژاد مهربان انجام شد. فراسنجههای گوارشپذیری و تعادل نیتروژن در 15 رأس بره نر نژاد مهربان در یک آزمایش 21 روزه شامل 16 روز عادتپذیری بـه شـرایط آزمـایش و 5 روز بـرای جمعآوری نمونهها انجام شد. جیرهها حاوی انرژی و پروتئین یکسان بودند. جیرههای غذایی شامل 1) کاه و کنجاله سویا، 2) کاه و کود اوره، 3) کاه و مکمل اوره آهستهرهش تجاری اپتیژن و 4) کاه و اوره آهستهرهش ساخته شده در آزمایشگاه بودند. قبل از انجام آزمایش درون تنی، نرخ و مقدار آزادسازی منابع اوره آهستهرهش در آب و ناپدید شدن شکمبهای اندازهگیری شد. گوارشپذیری ماده خشک و ماده آلی تحت تأثیر تیمارهای آزمایشی قرار نگرفت، در حالیکه گوارشپذیری الیاف نامحلول در شوینده خنثی و الیاف نامحلول در شوینده اسیدی در تیمارهای حاوی اوره آهستهرهش تفاوت معنیداری را نسبت به تیمار حاوی کاه و کنجاله سویا نشان دادند (05/0 ˂ P). مقدار نیتروژن دفع شده از راه ادرار در تیمار حاوی اوره به صورت معنیداری بالاتر بود، در حالیکه مقدار نیتروژن ابقا شده در جیرههای حاوی اوره آهستهرهش به صورت معنیداری (05/0 ˂ P) بیشتر از سایر تیمارها بود. نتایج این پژوهش نشان داد که اوره آهستهرهش تولید شده در این پژوهش دارای الگوی مناسب آزادسازی در شکمبه بوده و میتواند به عنوان یک محصول بومی در کشور در جیره نشخوارکنندگان حاوی علوفه نامرغوب مورد استفاده قرار گیرد.

اثر افزودنیهای مختلف بر تجزیه پذیری ماده خشک سیلاژ خردل علوفه ای (Brassica juncea) در مراحل مختلف فنولوژیک
پژوهش حاضر جهت بررسی میزان تجزیهپذیری گیاه خردل علوفهای (Brassica juncea)در مراحل مختلف رشد و اثر ملاس و اوره بر کیفیت و تجزیهپذیری ماده خشک سیلاژ آن، انجام شد.در مرحله اول، ترکیبات شیمیایی شامل ماده خشک و پروتئین خام در سه مرحله رشد فنولوژیک گیاه (رویشی، گلدهی و خمیری شدن دانه) اندازهگیری شد. سپس 5/1 کیلوگرم از گیاه در هر مرحله رشد، به قطعات 3 تا 5 سانتیمتری خرد و با ملاس و اوره مخلوط و در کیسههای پلاستیکی سیلو شد. تیمارهای اعمال شده در هر مرحله بر سیلاژها، شاهد، 5 درصد ملاس، 5/0 درصد اوره، 10 درصد ملاس، 1/0 درصد اوره و 5 درصد ملاس بهعلاوه 5/0 درصد اوره در نظر گرفته شد. سیلاژها به مدت 45 روز در دمای اتاق نگهداری شدند. پس از آن ماده خشک سیلاژها تعیین شد و مورد ارزشیابی ظاهری قرار گرفت و pH و دما اندازهگیری شد. در مرحله دوم فراسنجههای تجزیهپذیری ماده خشک گیاه و سیلاژ خردل با استفاده از روش کیسههای نایلونی تعیین شد.با نمو گیاه، ماده خشک گیاه خردل علوفهای بهطور معنیداری افزایش و پروتئین خام کاهش داشت. نتایج ارزشیابی ظاهری سیلاژها به وسیله نمرهگذاری نشان داد که سطوح مختلف ملاس سیلاژهایی با کیفیت بسیار خوب و خوب ایجاد کردند. در طی سیلو کردن، ماده خشک بهطور معنیداری کاهش یافت (05/0˂P) و در سیلاژهای عملآوری شده با ملاس، ماده خشک بهطور معنیداری بیشتر از سایر سیلاژها بود (05/0˂P). در سیلاژهای تهیه شده، pH بهوسیله ملاس، کاهش و با افزودن اوره، افزایش یافت. پایداری هوازی سیلاژهای مراحل رویشی و گلدهی بهطور معنیداری بیشتر از مرحله خمیری شدن دانه بود (05/0˂P). در هر سه مرحله رویشی، گلدهی و خمیری شدن دانه، بیشترین میزان درصد ناپدیدی ماده خشک، بخش سریع تجزیه، پتانسیل تجزیهپذیری و تجزیهپذیری موثر مربوط به سیلاژهای عملآوری شده با ملاس بوده و بیشترین میزان بخش کند تجزیه مربوط به سیلاژ بدون افزودنی بود.

اثر پولکی کردن با بخار با شرایط متفاوت بر تجزیهپذیری شکمبهای ماده خشک و پروتئین خام دانه جو در گاوهای هلشتاین
اثر پولکی کردن با بخار با شرایط مختلف، بر تجزیهپذیری شکمبهای ماده خشک و پروتئین خام دانه جو در گاوهای هلشتاین مورد بررسی قرار گرفت. این پژوهش به صورت آزمایش فاکتوریل در قالب طرح کاملاً تصادفی انجام شد. فرآوریهای مورد استفاده شامل دو سطح فشار بخار (3 و 5 بار)، سه سطح دما (75، 90 و 105 درجه سانتی گراد) و سه سطح فاصله غلتک (کم، متوسط، زیاد) بودند. مقادیر تجزیهپذیری با استفاده از روش کیسههای نایلونی با استفاده از سه رأس گاو هلشتاین فیستولادار تعیین شد. میزان تجزیهپذیری ماده خشک و پروتئین خام در اثر پولکی کردن با بخار در نمونه فرآوری شده در مقایسه با نمونه شاهد کاهش معنیداری نشان داد (05/0P<). افزایش فشار، دما و کاهش فاصله غلتک در اکثر ساعتهای انکوباسیون، سبب کاهش معنیدار تجزیهپذیری ماده خشک و پروتئین خام شد (05/0P<). در اثر فرآوری، میزان بخش تند تجزیه شونده (a)ماده خشک و پروتئین خام در نمونه فرآوری شده در مقایسه با شاهد کاهش یافت. کاهش معنیداری، در بخش کند تجریه شونده (b) ماده خشک و پروتئین نمونه فرآوری شده در مقایسه با نمونه شاهد مشاهده شد (05/0P<). علاوه بر اثر کاهشی فرآوری پولکی کردن با بخار بر میزان تجزیهپذیری موثر ماده خشک و پروتئین خام دانه جو، افزایش فشار، دما و کاهش فاصله غلتک به طور معنیداری میزان تجزیهپذیری موثر ماده خشک و پروتئین خام را در شکمبه کاهش داد (05/0P<). با توجه به اثر کاهنده پولکی کردن با بخار بر میزان تجزیهپذیری ماده خشک و پروتئین خام، این فرآیند میتواند خطر تجزیهپذیری بالای دانه جو و اسیدوز شکمبهای را کاهش دهد.

اثر سطوح مختلف اسانس مروتلخ بر عملکرد و برخی فراسنجههای خونی و ایمنی جوجههای گوشتی تحت تنش گرمایی
به منظور بررسی سطوح مختلف اسانس مروتلخ بر عملکرد و برخی فراسنجههای خونی و ایمنی جوجههای گوشتی تحت تنش گرمایی، آزمایشی در قالب طرح کاملاً تصادفی با چهار تیمار (صفر، 150، 300 و 450 میلیگرم در کیلوگرم اسانس مروتلخ)، به مدت 42 روز انجام شد. تنش گرمایی از سن 22 روزگی اعمال شد. هر سه سطح اسانس به کار برده شده باعث کاهش معنیدار مصرف خوراک در مقایسه با گروه شاهد در دوره آغازین شد، ولی در دوره رشد و کل دوره پرورش پرندگان تغذیه شده با سطح 450 میلیگرم در کیلوگرم اسانس بالاترین مصرف خوراک را نشان دادند. استفاده از هر سه سطح اسانس مروتلخ باعث کاهش معنیدار ضریب تبدیل خوراک در دورههای آغازین، رشد و کل دوره نسبت به تیمار شاهد شد (05/0P<). کاهش معنیدار کلسترول LDL و تریگلیسیرید خون با مصرف سطح 450 میلیگرم در کیلوگرم مشاهده شد (05/0P<). در سن 21 روزگی، افزودن اسانس باعث افزایش معنیدار لنفوسیت، مونوسیت شد و نسبت هتروفیل به لنفوسیت و تعداد هتروفیل کاهش معنیداری یافت (05/0P<). وزن بورس فابریسیوس و تیموس در سطح 450 میلیگرم در کیلوگرم اسانس افزایش یافت (05/0P<). وزن سنگدان و کل دستگاه گوارش با افزودن150 میلیگرم در کیلوگرم اسانس مروتلخ افزایش و وزن چربی حفره بطنی در سطح 450 میلیگرم در کیلوگرم کاهش معنیداری یافت (05/0P <). بیشترین درصد خاکستر استخوان درشتنی در سطح 450 میلیگرم در کیلوگرم اسانس مشاهده شد (05/0P<). با توجه به نتایج این پژوهش و بهبود عملکرد، کاربرد اسانس مروتلخ تا سطح 300 میلیگرم در کیلوگرم در شرایط تنش گرمایی در تغذیه جوجههای گوشتی توصیه میشود.

اثر سطوح مختلف تفاله انگور بر عملکرد تولید و کیفیت داخلی تخم مرغ در دما و زمان های مختلف نگهداری
آزمایشی به منظور بررسی اثرات سطوح مختلف تفاله انگور بر عملکرد مرغهای تخمگذار و ماندگاری کیفیت تخممرغ طی نگهداری در دما و زمانهای مختلف طراحی شد. تعداد 160 قطعه مرغ تخمگذار لگهورن بهصورت چهار تیمار با پنج تکرار و هشت مرغ در هر تکرار در قالب طرح کاملاً تصادفی برای سطوح صفر، 5/1، 3 و 5/4 درصد تفاله انگور استفاده و عملکرد تولید آنها به مدت هشت هفته بررسی شد. در پایان دوره، شش تخممرغ از هر تکرار مربوط به سطح 5/4 درصد تفاله، در دماهای 4 و 27 درجه سانتیگراد و زمانهای 1، 7 و 30 روز پس از تولید، نگهداری و کیفیت داخلی آنها بررسی شد. نتایج نشان داد که با تغذیه 5/4 درصد تفاله انگور به مدت هشت هفته، درصد تولید، وزن تخممرغ، توده تخممرغ و ضریب تبدیل خوراک برای تولید تخممرغ پرندگان تحت تاثیر قرار نگرفت، اگرچه مصرف خوراک کاهش یافت (05/0>P). نتایج آزمایش نگهداری تخممرغ در دما و زمانهای مختلف نیز نشان داد که با افزایش سطح تفاله به 5/4 درصد، pH سفیده کاهش و واحد هاو افزایش یافت. اثرات متقابل نیز نشان داد که 5/4 درصد تفاله انگور در جیره، توانست به طور معنیداری تغییرات pH سفیده و واحد هاو را در طول زمان نگهداری محدودتر کند. به طور کلی، میتوان تا 5/4 درصد تفاله انگور در جیره مرغهای تخمگذار در دوره پس از تولکبری اجباری بدون اثر منفی بر عملکرد استفاده کرد و خاصیت آنتیاکسیدانی تفاله انگور میتواند افت کیفی تخممرغ را در زمان نگهداری محدودترکند.

اثر منبع و اندازه ذرات فیبر جیره غذایی بر عملکرد و کیفیت تخممرغ در مرغ های تخمگذار تجاری
این تحقیق به منظور بررسی اثر منابع و اندازه ذرات فیبر بر عملکرد تولیدی و کیفیت تخممرغ در مرغهای تخمگذار انجام شد. آزمایش با 324 قطعه مرغ تخمگذار سویه لوهمن در سن 27 هفتگی با نه تیمار و شش تکرار به ازای هر تیمار به مدت 12 هفته انجام شد. تیمارهای آزمایشی عبارت بودند از: گروه شاهد، دو منبع فیبر (پوسته آفتابگردان و تفاله چغندرقند)، دو سطح فیبر (5/2 و 5 درصد) و دو اندازه ذرات فیبر (ریز، 2 میلیمتر و درشت، 6 میلیمتر). اثرات اصلی منابع فیبر، سطح فیبر و اندازه ذرات فیبر در کل دوره بر وزن تخممرغ، توده تخممرغ، درصد تولید تخممرغ، ضریب تبدیل غذایی، وزن مرغ و مصرف خوراک روزانه معنیدار نبود. به کار بردن 5 درصد پوسته آفتابگردان با اندازه ذرات درشت، وزن و ضخامت پوسته را در مقایسه با گروه کنترل در سن 30 هفتگی کاهش داد (05/0P< ). همچنین رنگ زرده در گروهی که 5/2 درصد پوسته آفتابگردان با اندازه ریز دریافت کردند کاهش و در 5 درصد تفاله چغندرقند، در مقایسه با شاهد افزایش یافت (0001/0P< ). واحد هاو در پوسته آفتابگردان نسبت به تفاله چغندرقند در سن 38 هفتگی کاهش یافت (05/0P< ). افزودن منابع فیبر، pH مدفوع مرغهای تخمگذار را نسبت به گروه شاهد افزایش داد (05/0P< ). رقیق کردن جیره با پوسته آفتابگردان و تفاله چغندرقند تا سطح 5 درصد اثر منفی بر عملکرد تولیدی مرغهای تخمگذار ندارد، در حالیکه باید اثرات آنها بر کیفیت تخممرغ مورد توجه قرار گیرد.
