اثر سطوح مختلف نانو ذرات اکسید روی و اکسید روی بر برخی فراسنجه‌های شکمبه‌ای بزغاله‌های نر مرغوز به روش برون‌تنی و درون‌تنی

چکیده

در این پژوهش اثر اکسید روی (ZnO) و نانو اکسید روی (nZnO) بر برخی فراسنجه‌های شکمبه در دو آزمایش برون‌تنی و درون‌تنی در قالب یک طرح کاملاً تصادفی بررسی شد. پنج تیمار آزمایشی شامل سطوح صفر (شاهد)، 20 و 40 قسمت در میلیون عنصر روی از منبع ZnO  و 20 و 40 قسمت در میلیون روی از منبع nZnO بود که به جیره پایه مکمل شد. در آزمایش برون‌تنی، فراسنجه‌های روند تخمیر شکمبه‌ای در طول 144 ساعت انکوباسیون متوالی (GP144) و حجم گاز تولید شده در طول 24 ساعت انکوباسیون (GP24) و فراسنجه‌های تخمیر مربوطه به‌طور جداگانه مورد بررسی قرار گرفتند. در آزمایش درون‌تنی، تعداد 20 رأس بزغاله نر مرغوز 8-7 ماهه با میانگین وزن 74/3± 67/17 کیلوگرم به‌مدت 14 روز از تیمارهای آزمایشی تغذیه شدند و pH، غلظت آمونیاک و کل اسیدهای چرب فرار و نیز جمعیت پروتوزوآها در مایع شکمبه تعیین شد. نتایج نشان داد GP144و GP24و فراسنجه‌های حاصل از آنها (به‌جز حداکثر ظرفیت تولید گاز (A) و فاز تأخیر (L)) و نیز فراسنجه‌های شکمبه‌ای در آزمایش درون‌تنی، تحت تأثیر مکمل روی قرار نگرفتند. اختلاف معنی‌داری در خصوص تعداد کل پروتوزوآها در بین تیمارها مشاهده نشد. بیشترین تعداد مربوط به جنس انتودینیوم و کمترین آن مربوط به جنس ایزوتریشا بود. به‌طورکلی استفاده از سطوح 20 و 40 قسمت در میلیون روی از طریق هر دو مکمل ZnO  و nZnO  بر فراسنجه‌های شکمبه‌ای به دو روش برون‌تنی و درون‌تنی اثر معنی‌داری نداشت.

In this study the effects of zinc oxide (ZnO) and zinc oxide nano particles (nZnO) were investigated on some parameters of rumen with either in vitro or in vivo experiments based on a completely randomized design. The experimental treatments (5 treatments) were included levels of zero (control), 20 and 40 ppm zinc as ZnO and 20 and 40 ppm zinc as nZnO  that were added to the basal diets. In in vitro experiments, rumen fermentation kinetics parameters during 144 h continuous incubation (GP144) and the volume of gas produced during the 24 h incubation (GP24) and the relevant fermentation parameters were analyzed separately. In in vivo experiments, 20 male Markhoz goat kids aged 7-8 months were fed for 14 days with experimental treatments and the pH, ammonia and total volatile fatty acids concentration and protozoa populations in rumen fluid were determined. Results showed, GP144 and GP24 and the relevant parameters (except for asymptotic gas production (A) and lag time (L)) and rumen parameters in in vivo experiment were not affected by zinc sources. No significant difference was observed between treatments for total number of protozoa. Highest number was seen for Entodinium spp. and lowest for Isotricha spp. . Overall, the levels of 20 and 40 ppm zinc as ZnO and nZnO had no effect on rumen parameters by either in vitro or in vivo methods.
